A simple technique for the preparation of polytene chromosomes from Culex quinquefasciatus.
نویسنده
چکیده
Preparation of polytene chromosomes in Culex species is very difficult and the available techniques are not always reproducible. Presuming these problems might be due to lack of an appropriate technique to deal with these long and fragile chromosomes invested with ectopic pairings and sticky ends, certain promising methods with several modifications have been tried. During the process, recently, a simple and reproducible method was developed for obtaining good polytene chromosomes from Culex quinquefasciatus. The detailed methodology with precautions and its advantages over other reported techniques are described.

Malpighian tubule polytene chromosomes of Culex quinquefasciatus (Diptera, Culicinae).
Dipteran polytene chromosomes provide an excellent model for understanding in species complexes, as well as for structural and functional cytogenetics. The status of species in the Culex pipiens complex is controversial and the use of polytene chromosomes for cytogenetic analysis in the subfamily Culicinae has been difficult because of methodological problems. In this study, Malpighian tubule p...
متن کاملFluorescent in situ Hybridization on Mitotic Chromosomes of Mosquitoes
Fluorescent in situ hybridization (FISH) is a technique routinely used by many laboratories to determine the chromosomal position of DNA and RNA probes. One important application of this method is the development of high-quality physical maps useful for improving the genome assemblies for various organisms. The natural banding pattern of polytene and mitotic chromosomes provides guidance for th...
متن کاملMitotic-Chromosome-Based Physical Mapping of the Culex quinquefasciatus Genome
The genome assembly of southern house mosquito Cx. quinquefasciatus is represented by a high number of supercontigs with no order or orientation on the chromosomes. Although cytogenetic maps for the polytene chromosomes of this mosquito have been developed, their utilization for the genome mapping remains difficult because of the low number of high-quality spreads in chromosome preparations. Th...
متن کاملLarvicidal potential of Cyathea species against Culex quinquefasciatus
Resistance to insecticides has persuaded researchers to find new methods to control Culex quinquefasciatus proliferation. Plants may be a source of alternative agents for mosquito control due to ever-growing insecticide resistance in mosquito vectors and environmental imbalance caused by synthetic insecticides. The present study was intended to study the larvicidal activity of selected Cyathea ...
متن کاملToxic Response of Mosquito Culex Quinquefasciatus (Diptera: Culicidae) to some Agricultural Pesticides (Butachlor and Pertilachlor)
Background: The expansion of herbicide used in aquatic ecosystems as well as in terrestrial if is not properly controlled may produce harmful effects on freshwater fisheries. Residue limits of these agricultural chemicals in tropical fishery waters should be established. The aim of this study was to determine the acute toxicity of butachlor and pertilachlor as potential dangerous herbicides to ...
